The invention relates to a method for marker-free examination of physical, chemical and/or biochemical reactions, bonding and addition or other interactions in/or on a thin layer. According to said method the detection of a range consisting of several wavelengths, as carried out in a known manner, on the base of reflection measuring is reduced to the detection of one wavelength. The measurement of the intensity modification of a light fraction reflected on the thin layer taken place as a result of interactions in/or on the thin layer is carried out for one wavelength in order to evaluate the measurement results. Said wavelength preferably corresponds to a wavelength whose light intensity modification is maximum after a reaction or interaction carried out on the thin layer. The optimal measuring wavelength depends on the thin layer properties, in particular on the optical thickness thereof and the properties of a carrier to which said thin layer is applicable and on the angle of incidence and polarisation of incident light on the thin layer. A device for carrying out said method is also disclosed.
