An immunoblotting kit for detecting a major antigen of Pleurotus florida worker lung is provided to perform a first selective diagnosis of hypersensitivity pneumonitis caused by Pleurotus florida basidiospore, and search the major antigen from the Pleurotus florida basidiospore, thereby developing an accurate diagnosing method about the Pleurotus florida worker lung. The immunoblotting kit for detecting a major antigen of Pleurotus florida worker lung comprises at least one antigen isolated from Pleurotus florida basidiospore and selected from the group consisting of antigen having a molecular weight of 140kDa, antigen having a molecular weight of 78kDa, and antigen having a molecular weight of 62-53kDa, human serum, and a conjugate solution. In the kit, the concentration of the Pleurotus florida antigen is 10-20 mug/ml.
