A novel glycosylase isolated from a thermophilic microorganism is provided to diagnose genetic diseases rapidly and accurately by detecting gene disorders such as adenine/oxoguanine mispair in a living body without side effects. A novel glycosylase isolated from Thermus thermophilus(ATCC 33923) has the recognition activity of thymine/oxoguanine mispair and adenine/oxoguanine mispair and comprises a part of or the total of the amino acid sequence set forth in SEQ ID NO:1. A gene encoding glycosylase which acts on the adenine/oxoguanine mispair comprises a part of or the total of the nucleotide sequence set forth in SEQ ID NO:2. An Escherichia coli expression vector pMALc2xthr-TTC1535 contains the adenine/oxoguanine mispair glycosylase gene. A transformed Escherichia coli TB1(pMALc2xthr-TTC1535) is produced by transforming Escherichia coli with the Escherichia coli expression vector pMALc2xthr-TTC1535.
