A method for preparing a human milk reinforcing agent for premature infants is provided to sufficiently supply nutritional elements required for the growth of premature infants while not increasing the nursing amount. The method for preparing a human milk reinforcing agent for premature infants comprises the steps of: mixing 6-7.5 wt% of dextrin, 0.03-0.07 wt% of taurine, 1-2 wt% of trace minerals and 0.57-1.43 wt% of vitamins to form a first mixture; mixing powdery milk for premature infants containing 10-15 wt% of mixed edible oil and fats beta containing Sn-2 palmitic acid, 15-25 wt% of dextrin and 5-10 wt% of MCT oil, 8.4-10.5 wt% of high-protein whey powder, 21-25 wt% of defatted milk, 9-12 wt% of lactose, 6-7.5 wt% of dextrin and 11-12 wt% of minerals other than the minerals used in the first mixture to form a second mixture; and further mixing the first mixture with the second mixture.
