A porcine islet cell isolation method for xenotransplantation is provided to obtain a sufficient amount of porcine islet cells with high purity stably and efficiently, and to help treatment of diabetes through transplantation of islet cells. The porcine islet cell isolation method for xenotransplantation comprises the steps of: extracting the pancreas from bacteria free pigs; introducing solution containing a specific enzyme into the extracted pancreas; collecting the islet cells released from the pancreas by decomposing the fibrous tissue of the pancreas with the enzyme; purifying the collected islet cells, wherein the specific enzyme is liberase PI; the solution containing the specific enzyme is prepared by mixing buffer II containing water, raffinose, histidine, lactobionic acid and electrolyte with insulin, and adding liberase PI; the released islet cells are collected at 28-29 deg.C with a Ricordi collector; the concentration of liberase PI is 0.75 to 1.5 mg/ml and the amount of solution introduced is 1.7ml per 1g of the pancreas; and the pig is a 7 day or less old pig.
