Preparing docetaxel (III) from paclitaxel (I) comprises: (1) deacetylating (I); (2) protecting the free hydroxy groups in 7, 10 and 2' positions; (3) debenzoylating 3'-position of a primary amine derived from 10-deacetylbaccatin containing protected hydroxy groups in 7, 10 and 2' positions; (4) functionalizing amine with t-butoxycarbonyl radical to obtain a docetaxel derivative (II); and (5) releasing protected hydroxy groups to obtain (III). Preparing docetaxel (III) from paclitaxel (I) comprises: (1) deacetylating (I); (2) protecting the free hydroxy groups in 7, 10 and 2' positions; (3) debenzoylating 3'-position of a primary amine derived from 10-deacetylbaccatin containing protected hydroxy groups in 7, 10 and 2' positions; (4) functionalizing amine with t-butoxycarbonyl radical to obtain a docetaxel derivative of formula (II); and (5) releasing protected hydroxy groups to obtain (III).
