The invention relates to pharmaceuticals industry, in particular to the technology of preparation of medicinal agents of clam tissues, and can be used as medicinal agent for functional recovery of human surfactant system. A method for the preparation of surface-active composition (Molufil trademark) for recovery of lung surfactant function at surfactant deficiency involves comminution of tissues of mussel mollusks as raw material in frozen state, extraction of raw material with ethanol, separation of ethanol extract from raw material residues, reextraction of the obtained residue with ethanol acidified with acetic acid to pH 4-5 followed by combination of ethanol extracts after the first and second extraction, removal of ethanol by vacuum stripping, cleaning of the obtained fraction from hydrocarbon derivatives, fatty acid esters, cholesterol oxidation products and esters thereof with further resuspending of the obtained lipid-protein fraction in ethanol with addition of hexane and ethyl acetate mixture in ratio of 10:20:1 and separation of hexane-ethyl acetate layer from water-alcohol fraction, boiling-down of the obtained extract to1/10-1/100 of volume after extraction, cleaning of the obtained fraction with activated carbon with further filtration of solution from residues of protein components by means of membrane filters "Milipor 0,2", concentration of purified and clarified filtrate by boiling and addition of vitamin E in concentration of at least 0.05% and preparation of substance from which 1% alcohol solution which is suspended with physiological salt solution and heparin is prepared. The suspension obtained is packed to 10 ml sterile flasks, lyophilized, and surface-active composition Molufil is obtained as white powder with yield of 5%, this is used for recovery of surfactant deficiency.
