A cross-linked poly(vinyl alcohol)-collagen hydrogel scaffold for cell culture and a preparation method thereof are provided to have good physical characteristics such as gel strength. A preparation method of cross-linked poly(vinyl alcohol)-collagen hydrogel scaffold for cell culture includes the steps of: preparing collagen IV aqueous solution by water-bath collagen IV; preparing PVA aqueous solution by water-bathing PVA; preparing PVA/collagen IV aqueous solution by mixing the PVA aqueous solution and collagen aqueous solution prepared in the aforesaid steps in the ratios of 7:3 to 9:1 based on the weight of solid thereof; and applying radiation at 25 to 50kGy to the PVA/collagen IV aqueous solution.
