An adult stem cell derived from human cord blood is provided to grow cells vigorously for a long time during an indifferentiation step compared with a conventional adult stem cell by being coupled to an osteoclast based niche-like structure and have the capability of being differentiated into various kinds of cells such as neurons, bone forming cells and fat cells, thereby being usefully used for treating an incurable diseases, nervous system diseases and skeletal system diseases. A method for preparing an adult stem cells comprises a step of culturing monocytes isolated from human cord blood in a DMEM medium containing 1-30% FBS and bFGF under the condition of 8-12% CO2, wherein the adult stem cell is characterized in that it shows positive immunological characteristics on CD29, CD44, CD31 and CD105 and negative immunological characteristics on CD4, CD20, CD34 and CD51, shows positive immunological characteristics on Oct4, has a structure where round shaped stem cells are contacted on osteoclasts surrounded by spindle-shaped cells attached to a plastic bottom, and has the capability of being differentiated into cells derived from an endoderm, an ectoderm and a mesoderm.
