This invention relates to a novel media composition for the growth of genetically manipulated Rhizobia comprising K2HPO4 0.3 to 0.7 g/litre preferably 0.5g; mustard cake 0.4 to 1.5g/litre preferably 0.6g, Molasses 0.3 to 0.9g/litre preferably 0.6g and yeast extract 0.3 to 0.6g per litre preferably 0.3g having pH 6.8 and the process of efficient production of Poly hydroxy butyrate towards cost effective biodegradable plastics using Rhizobia from Agri-byproducts and the Rhizobial mode of preparation of Polyhydroxy butyrate comprising the steps of growing Rhizobia in the above mentioned media at 27°C to 30°C preferably at 28 °C under stationary conditions for one day, harvesting cell biomass by centrifugation at 6000 rpm, treating the cell pellet slurry with 10-20 ml of each of Sodium hypochlorite and chloroform per gm of slurry and evaporating/re-cycling the chloroform leading to dry Poly hydroxy butyrate powder.
