A large volume stacking method using an electro-osmotic flow pump in capillary electrophoresis electrospray ionization mass spectrometry and an apparatus for the same are provided to supply run buffer in opposite direction to the sample injection by temporarily installing a run buffer storage container in a capillary tube outlet on the orifice side of the mass spectrograph while sample is accumulated. A large volume stacking method using an electro-osmotic flow pump in capillary electrophoresis electrospray ionization mass spectrometry comprises step of: injecting the sample with liquid of lower conductivity than a run buffer as matrix into a capillary tube; putting the run buffer in the inlet part of the capillary tube; that the capillary tube outlet is immersed; starting the accumulation of sample by applying reverse voltage in the inlet part of the capillary tube; removing the outlet reservoir when the current of the capillary electrophoresis abruptly changes; and giving sheath solution in the mass spectrograph and applying the electrospray voltage so that concentrated sample moves to the mass spectrograph.
