A method of manufacturing technetium-99m cover iron oxide nanoparticle is provided to inject iron oxide nanoparticles easily into a body using acid solution or borohydride anion exchange resin as a reducing agent and to grasp easily particle distribution location and exhaustion course to vitro etc. A method of manufacturing technetium-99m cover iron oxide nanoparticle indicated as a chemical formula 1 comprises a step of reacting ferric oxide(Fe2O3) with sodium pertchenetate(Na99mTcO4) under acid solvent. In the chemical formula 1, M is alkali metal. The acid catalyst is one selected from a group consisting of hydrochloric acid, nitric acid and sulfuric acid. The reaction is performed in 98 ~ 100 °C. The reaction time is 20 ~ 40 minutes.
