A method for forming spore of antifungal polyene generating actinomycetes is provided to optimize the conversion efficiency to spore under the condition of sonication and produce the spore having high concentration. A method for forming a spore of antifungal polyene generating actinomycetes comprises: a step of culturing antifungal polyene generating actinomycetes; a step of pulverizing by sonication; and a step of collecting changed spore. The antifungal polyene generating actinomyces is Pseudonocardia autotrophica or Streptomyces sp.
