DNA POLYMORPHISM IDENTITY DETERMINATION USING FLOW CYTOMETRY

PPROBLEM TO BE SOLVED: To provide a sensitive, uniform and flexible method for determining the DNA base composition in a specific site. PSOLUTION: In the method, primers designed to be immobilized on microspheres are allowed to anneal to the DNA strand under investigation, and are extended by either DNA polymerase using fluorescent deoxynucleotides or ligated by DNA ligase to fluorescent reporter oligonucleotides. The deoxynucleotide or reporter oligonucleotide attached to the immobilized primer is measured by flow cytometry, thereby the nucleotide polymorphism on the DNA strand is identified.
