An extraction method of patinopecten yessoensis polysaccharide comprises process steps of raw material treatment, homogenization, supersonic treatment, enzymolysis, separation, concentration, precipitation with alcohol, and drying. As for said raw material treatment, patinopecten yessoensis meat tissue is directly homogenized by adding water or homogenized by adding water after being oven dried or freeze dried; said enzymolysis is preformed with one or two of pepsin, trypsase, Bacillus subtilis neutral proteinase. Said meat tissue is scallop meat, scraps other than scallop meat, partial scraps, rod shaped scallop meat and partial scraps or all meat tissue other than shell. There is provided a process of extracting polysaccharide from patinopecten yessoensis, particularly from patinopecten yessoensis scraps with taking full advantage of scraps. The product obtained has high purity, and high yield. The patinopecten yessoensis polysaccharide produced can be used as either a single product or a base material which is combined with other food to give various multifunctional food.
