PPROBLEM TO BE SOLVED: To provide a method and a device for accurately detecting plasmodium in high accuracy with eliminating effects of reticulocytes independent of species of samples and obtain a reagent kit for the same. PSOLUTION: The method for detecting plasmodium is to prepare a sample for measurement by isolating the plasmodium by hemolyzing erythrocytes in a test sample and dyeing the isolated plasmodium with a fluorescent pigment, detect optical information from the test sample and detect the plasmodium by discriminating the same depending on the optical information. The method for detecting the plasmodium is to treat the sample with a first hemolytic agent containing a first surfactant and treat the treated sample with a second hemolytic agent containing a second surfactant having hemolytic action stronger than that of the first surfactant.
