METHOD OF MANUFACTURING AN AMORPHOUS FORM OF THE HEMI-CALCIUM SALT OF (3R, 5R) 7- 3-PHENYL-4-PHENYLCARBAMOYL-2-(4-FLUOROPHENYL)-5-ISOPROPYL-PYRROL-1-YL-3, 5-DIHYDROXYHEPTANOIC ACID (ATORVASTATIN)
In the present invention, there is disclosed a process for preparing amorphous form of hemicalcium salt of (3R,5R)7-[3-phenyl-phenylcarbamoyl-2-(4-fluorophenyl)-5-isopropylpyrrol-l-yl]-3,5-dihydroxyheptanoic acid of the general formula I, in which process the (3R,5R) tert-butyl (6-{2-[3-phenyl-4-phenylcarbamoyl-2-(4-fluorophenyl)-5-isopropylpyrrol-1-yl]ethyl}-2,2-dimethyl-[1,3]dioxan-4-yl)-acetate is converted by acid and subsequently alkali hydrolysis to a salt of the (3R,5R) 7-[3-phenyl-4-phenylcarbamoyl-2-(4-fluorophenyl)-5-isopropylpyrrol-1-yl]-3,5-dihydroxyheptanoic acid with an alkali metal wherein the obtained acid is then extracted after optional acidification into an organic solvent being selected from the group consisting of esters of an acid containing 1 to 5 carbon atoms with an alcohol having 1 to 5 carbon atoms. After purification operation are carried out the co the compound is then converted by the treatment with a calcium salt or calcium hydroxide, or calcium alcoholate containing 1 to 5 carbon atoms to a hemicalcium salt, which is then precipitated with a hydrocarbon having 5 to 1 carbon atoms or with a dialkylether of the general formula Ri1ORi2 wherein Ri1 and Ri2 represent an alkyl group containing 1 to 5 carbon atoms.
