The invention relates to methods for measuring the intracellular quantity of an intrinsically fluorescent molecule of interest in cells, comprising the following steps consisting in: measuring the fluorescence of the cells for which the intracellular quantity of the molecule is required, using a flow cytometer parameterised with the emission and absorption spectra of the molecule of interest, said fluorescence value being plotted on a line of correlation between the fluorescence intensity and the intracellular concentration of the molecule of interest; and reading the intracellular quantity of the molecule of interest.
