The method involves providing a biological sample structure that is labeled with a substance. A molecule of the substance is present in two states (A, B) such as fluorescent and non-fluorescent states, where the states differ from one another in photo physical property. The substance is used such that there is high probability that portions of the molecule will be in the respective states. Change of state of the molecule occurs spontaneously between the two states in both directions. A dwell time of the molecule in the states is influenced by change of an external parameter e.g. temperature.
