PURPOSE: A method for producing an outer membrane vesicle for multi-valent epitope delivery using a recombinant G(-) bacteria is provided to ensure safe outer membrane vesicle and use as a bio-nano vaccine. CONSTITUTION: A method for producing a recombinant G(-) bacteria which is able to fuse outer antigen with outer membrane protein comprises: a step of producing G(-) bacteria mutant strain in which a gene relating to lipid A biosynthesis of lipopolysaccharide(LPS) is delete; a step of producing the recombinant to fuse and express outer membrane protein an outer epitope; and a step of introducing the recombinant to the G(-) bacteria mutant strain. The gene relating to the lipid A synthesis of LPS is lpxL(htrB), lpxM(msbB) or pagP. A method for producing an outer membrane vesicle comprises: a step of producing G(-) bacteria mutant strain in which the gene relating to lipid A synthesis of LPS; a step of collecting outer membrane vesicle isolated from the G(-) bacteria mutant strain; and a step of inserting a synthetic peptide DNA fragment which encodes the synthetic peptide into internal space of the outer membrane vesicle.
