PURPOSE: A bio disc having a multi-layer diagnostic channel and a measuring method thereof are provided to effectively detect the diagnostic channel of a multi-layer structure by realizing the diagnostic channel for a lot of inspection items on one bio disc.;CONSTITUTION: A bio disc having a multi-layer diagnostic channel comprises a diagnosis area and a record area. The diagnosis area includes a diagnosis chamber(11) for analyzing specimen. The diagnosis area comprises at least two layers of diagnosis channel(12). Data read and data write are prevented from the diagnosis area. The record area is formed so that writing and reading the data required for the diagnosis of other area than the diagnosis area are possible. The at least two layers of diagnosis channel are overlapped each other. Respective reflecting layer(14) is arranged in the at least two layers of diagnosis channel.
