PURPOSE: Bio specimen measuring device and method using the evanescent wave of a prism are provided to enable an uniform and highly sensitive fluorescence signal to be measured on wider measuring region by the construction of exiting fluorescent material using a specially manufactured prism to be able to make total internal reflection.;CONSTITUTION: A bio specimen measuring device using the evanescent wave of a prism comprises a light source(2), a prism(3), a specimen(4), a detector(5) and a controller(6). The light source supplies the excited light. The prism is formed around the light source and receives the excited light(7). The specimen is formed around the prism. The specimen comprises a bio specimen receiving the evanescent wave(8) from the prism. The detector is formed around the specimen and detects the fluorescent light(9) generated from the specimen.
