The invention relates to a method for measuring the activity of nitricoxide synthase and a corresponding method for identifying nitric oxidesynthasemodulators wherein the method is a method of measuring a nitric oxide (NO)synthase activity, comprising the steps of: (a) incubating NO synthase inincubation medium with labeled arginine as substrate in a reaction mixturecontained in a reaction vessel, whereby arginine is converted into citrullineby theNO synthase, (b) separating nonconverted arginine from the reaction mixtureandcapturing non-converted arginine by means of a filter plate membrane; and(c) measuring the amount of captured arginine on the filter plate membrane asanindication of the NO synthase activity.
