PURPOSE: A mutant Nanoarchaeum equitans Neq A523R DNA polymerase is provided to improve processivity and extension rate and to enhance specificity and amplification efficiency under the presence of dUTP.;CONSTITUTION: A mutant Nanoarchaeum equitans Neq A523R DNA polymerase has an amino acid sequence of sequence number 2 in which 523th amino acid, alanine is substituted with arginine. A gene having a nucleic acid sequence encoding the polymerase has a sequence of sequence number 3. A recombinant vector contains the gene. The recombinant vector is Penpa523r. An E.coli transformed by the recombinant vector is Escherichia coli BL21-CodonPlus(DE3)-RIL/pENPA523R(deposit number: KCCM11031P). A method for preparing the polymerase comprises: a step of preparing the recombinant vector; a step of inserting the recombinant vector into a host cell for transformation; a step of culturing the transformant; and a step of collecting the polymerase from the transformant.;COPYRIGHT KIPO 2011
