The present invention relates to the method of synthesis of core-shell nanomedicine serving as a novel platform for the encapsulation of multiple therapeutic molecules enabling combinatorial therapy against diseases including cancer, inflammatory and auto-immune diseases and its associated manifestations. The core-shell nano-construct comprises of a biodegradable and biocompatible polymer as the core and another polymer or a protein as the shell, aiding following constructs (i) polymer core-polymer shell and (ii) polymer core-protein shell. The core-shell nanomedicine is developed in a manner to encapsulate at least one anti-cancer agent each in both the core and shell. Besides enhancing solubility, stability and bioavailability of the drugs, the core-shell platform aims at improving the therapeutic index by aiding sequential/simultaneous delivery of at least two different anti- drugs at the disease site, in combination. In addition, this core-shell nanomedicine can be targeted specifically to the preferred site of action by suitably surface conjugating the nano-construct with active biomolecules such as monoclonal antibodies, peptides, sugar moieties, vitamins etc.
