METHODS OF TREATING VASCULAR DISEASES CHARACTERIZED BY NITRIC OXIDE INSUFFICIENCY

The present invention provides methods of treating and/or preventing vasculardiseases where nitric oxide insufficiency is a contributing factor byadministering a therapeutically effective amount of at least one antioxidant,or a pharmaceutically acceptable salt thereof, and at least one of isosorbidedinitrate and isosorbide mononitrate, and, optionally, at least one nitrosatedangiotensin-converting enzyme inhibitor, nitrosated beta-adrenegic blocker,nitrosated calcium channel blocker, nitrosated endothelin antagonist,nitrosated angioetnsin II receptor antagonist, nitrosated renin inhibitor,and/or at least one compound used to treat cardiovascular diseases. Theantioxidant is preferably a hydralazine compound or a pharmaceuticallyacceptable salt thereof. The present invention also provides methods oftreating and/or preventing vascular diseases where nitric oxide insufficiencyis a contributing factor by administering a therapeutically effective amountof at least one nitrosated angiotensin-converting enzyme inhibitor, nitrosatedbeta-adrenergic blocker, nitrosated calcium channel blocker, nitrosatedendothelin antagonist, nitrosated angiotensin II receptor antagonist,nitrosated renin inhibitor, and, optionally, at least one antioxidant and/orat least one compound used to treat cardiovascular diseases. The presentinvention also provides methods of treating and/or preventing Raynaud'ssyndrome by administering a therapeutically effective amount of at least oneantioxidant and at least one of isosorbide dinitrate and isosorbidemononitrate and/or at least one nitrosated angiotensin-converting enzymeinhibitor, nitrosated alcium channel blocker, nitrosated endothelinantagonist, nitrosated angiotensin II receptor antagonist nitrosated renininhibitor. The present invention also provides novel transdermal patchescomprising at least one antioxydant and at least one of isosorbide dinitrateand isosorbide mononitrate and/or at least one nitrosated angiotensin-converting enzyme inhibitor, nitrosated beta-adrenergic blocker, nitrosatedcalcium channel blocker, nitrosated endothelin antagonist, nitrosatedangiotensin II receptor antagonist, nitrosated renin inhibitor.
