Disclosed in the present invention is a method for detecting pyrazinamide resistance of a Mycobacterium tuberculosis, comprising the steps of: A. splitting the Mycobacterium tuberculosis to be a template of a PCR reaction; B. designing a primer to amplify a full-length pyrazinamide enzyme gene; C. concentrating the amplified pncA gene fragments and performing a quantification; D. expressing the pncA enzyme using a wheat germ cell-free expression system; E. determining the activity of the expressed pncA enzyme and making a comparison with the pncA enzyme activities of the Mycobacterium tuberculosis standard sensitive strain and resistant strain so as to determine the pyrazinamide resistance of the Mycobacterium tuberculosis.
