PPROBLEM TO BE SOLVED: To provide efficient methods for producing cytidine-5'-monophosphate-N-acetylneuraminic acid and an N-acetylneuraminic acid-containing saccharide. PSOLUTION: This method for producing cytidine-5'-monophosphate-N-acetylneuraminic acid (CMP-NeuAc) is characterized by using a culture product of a microorganism having an ability for producing a protein selected from the below-described (1) and (2) or a treated product of the culture product as an enzyme source, making the enzyme source, CTP and N-acetylneuraminic acid to exist in an aqueous medium, producing and accumulating the CMP-N-acetylneuraminic acid (CMP-NeuAc), and then collecting the CMP-NeuAc from the aqueous medium. (1) A CMP-NeuAc synthase having a specific amino acid sequence. (2) A protein encoded in a DNA hybridized with a DNA having a base sequence complementary to a specific base sequence under stringent conditions and having a CMP-NeuAc synthase activity.
