Present investigation deals with development of novel small interfering RNA nanoconstructs for the chemosensitization of Gemcitabine. Lipid based non-viral delivery vectors can be utilized to develop siRNA encapsulated nanoconstructs. Ribonucleotide Reductase subunit-1 (RRM1), a gene responsible for chemoresistance of Gemcitabine, can be targeted using RRMlsiRNA to suppress the expression of this gene. In-vitro cell line evaluation in lung carcinoma cell lines can be utilized to confirm the efficacy of prepared nanoconstructs. In-vivo studies in T-cell suppressed mice can be performed to assess the performance of prepared nanoconstructs.
