CAPTURE PROBES AND PRIMERS FOR GENOTYPING OF CHRONIC MYELOID LEUKEMIA, AND USES THEREOF

PURPOSE: A method for detecting a chronic myelogenous leukemia fusion genotype using a capture probe and a primer is provided to remarkably improve sensitivity, to shorten detection time, and to automate detection steps by simultaneously performing reverse transcriptase (RT)-PCR, asymmetric PCR, and hybridization. CONSTITUTION: A capture probe for genotyping of chronic myelogenous leukemia contains base sequences selected from a group consisting of sequence numbers 6, 7, 8, 9, 11, and 12. A primer for genotyping of chronic myelogenous leukemia contains base sequences selected from the group consisting of sequence numbers 2-4. A method for detecting a chronic myelogenous leukemia fusion genotype comprises the steps of: crushing cells extracted from bone marrow or peripheral blood and acquiring a cell extract; mixing the cell extract, a primer set, DNA polymerase, reverse transcriptase, dNTP (dATP, dCTP, dTTP, dGTP), and a buffer solution and preparing a reaction mixture; transferring the reaction mixture onto the surface of a solid support on which a capture probe for genotyping is fixed; performing RT-PCR using RNA to prepare cDNA; amplifying BCR-ABL gene of cDNA by asymmetric PCR; denaturing the amplified gene; hybridizing the denatured gene with the capture probe and promoting the formation of a branched nucleic acid complex; and detecting the branched nucleic acid complex.
