PROBLEM TO BE SOLVED: To provide methods useful in constructing libraries that collectively display members of diverse families of peptides, polypeptides or proteins and the libraries produced using those methods.SOLUTION: Provided is a library encoded by a plurality of nucleic acids available through a method comprising the steps of: (i) amplifying the nucleic acids using a primer; (ii) rendering the amplified nucleic acids single-stranded; (iii) contacting the amplified single-stranded nucleic acids with a single-stranded oligonucleotide; and (iv) cleaving the nucleic acids at a restriction endonuclease recognition site; wherein the contacting and the cleaving steps are performed at a temperature, wherein the single-stranded nucleic acids are associated with the single-stranded oligonucleotide to form a partially double-stranded region of the single-stranded nucleic acids, wherein the remaining part of the single-stranded nucleic acids is single-stranded, and wherein the restriction endonuclease is active at the temperature.
