The present invention discloses a method of production of chondroitin sulfate with an average molecular weight (Mw) of 10-30 kDa by chemical sulfation, starting from the main chain of unsulfated chondroitin, obtained in turn by acid hydrolysis of the capsular polysaccharide K4, obtained directly from E. coli strain O5: K4: H4 or directly produced from a genetically modified E. coli strain. Sulfation of the N-acetyl-D-galactosamine residue at position 4 or 6 takes place simultaneously in the same polysaccharide chain, mimicking the character of sulfation observed in natural chondroitin sulfate, in contrast to sulfation obtained by the synthesis methods described to date.
