The present invention provides a process for generating 3D tumour models and tumoroids for drug screening and testing. The process includes the step of preparation of chitosan- gelatin (CG) scaffolds. The CG scaffolds are prepared by freeze drying method in which chitosan and gelatin are dissolved in a solvent, and are allowed to cross link in presence of a cross-linker, followed by lyophilising of the solution to form a porous hydrogel. The step of preparation of CG scaffolds is followed by sterilisation of CG scaffolds and seeding of cancer cells on these scaffolds. Thereafter, the cancer cell lines are allowed to adhere and grow on the scaffolds to allow for tumoroid formation. Thereafter, the 3D tumour models are tested for their characteristics in comparison with 2- dimensional (2D) tumour cell cultures and tumours grown in vivo. It is found that the 3D tumour models closely resembles in vivq tumour growth, which was not been achieved by 2D tumour cell culture. The 3D tumour model is further tested with NCl-H460 (Non-Small Cell Lung Cancer- NSCLC), HeLa (cervical cancer), MCF-7 (breast cancer), MG-63 (osteosarcoma), PaCa (pancreatic cell line), NT8e (human head and neck cancer), SF268 (glioblastoma cell line), and U87 (glioblastoma cell line) and analysed for its potential as a universal scaffold for generation of tumoroid-like structures.
