Calcium sensors and methods for detecting intracellular free calcium. # The invention provides a system based on a fusion protein comprising (i) a first polypeptide domain capable of binding Ca {sup, 2 +} sensor calcium, (ii ) a second polypeptide domain of SEQ ID NO: 1 (GFPuv) or a variant thereof which maintains two maxima in its excitation spectrum and at least one peak in its emission spectrum, and (iii) a flexible peptide connects the first domain and the second domain covalently.
