Determining an analyte in a sample, comprises: providing a test element; applying the sample to the test element; and determining the presence and/or amount of the analyte on the test element. The test element comprises an analyte-binding receptor molecule. A ligand-binding domain of the receptor molecule is present in a native, truncated, or mutated form and is optionally conjugated with a heterologous molecule. Independent claims are included for: (1) a test element for determining an analyte, comprising a first portion adapted to receive eluent, a second region formed for applying the sample containing the analyte, a third region formed for the optical detection of the analyte, a fourth region adapted to receive excess eluent, and a housing; (2) a sampling member for receiving an analyte of an object and for transmission of the analyte to a test element; and (3) a kit for determining an analyte, comprising a test element and a sampling member.
