The present invention relates to a biomarker composition comprising mitochondrial calcium uniporter (MCU) for diagnosing resistance to bortezomib used as a therapeutic drug against multiple myeloma and a diagnostic kit using the biomarker composition. The present invention provides a biomarker composition for diagnosing bortezomib resistance, comprising MCU gene or a protein expressed from the gene, or an antibody specific to the protein and a kit for diagnosing bortezomib resistance comprising the same. Also, the present invention provides a method for diagnosing bortezomib resistance, comprising a step for measuring mRNA expression levels of the MCU gene or expression levels of a protein coded by the MCU gene from sample of a patient with multiple myeloma and comparing the levels with a normal control sample.
