The present invention relates generally to the detection and differentiation of Mycobacterium tuberculosis complex vis-a-vis Non-tuberculosis mycobacterium. More particularly, the present invention relates to a primer(s) and probe(s) sequences set capable of detecting specific nucleotide sequences of Mycobacterium tuberculosis and which eventually helps in differentiating Mycobacterium tuberculosis complex(MTBC) from other mycobacteria. The Invention also extends to provide a highly specific and sensitive kit for the detection of Mycobacterium tuberculosis complex and capable of differentiating the presence of MTBC with respect to other mycobacteria(s) present in a sample. In another aspect of the Invention, there is provided a method for detecting Mycobacterium tuberculosis and Non-tuberculosis mycobacterium by utilizing an amplification of the nucleic acid and a detection system thereof.
