Disclosed is a human Tim-3-Ig fusion protein capable of blocking a Tim-3 signaling pathway. The Ig fusion protein comprises a Tim-3 protein, a human immunoglobulin Ig fragment and a linker sequence of the two. According to the present invention, a human Tim-3-Ig gene is obtained by an artificial synthetic method, an expression vector containing the Tim-3-Ig gene is constructed, and the obtained Ig fusion protein is subjected to an expression verification experiment, a binding activity experiment, blocking activity experiments in different cell lines and an in vivo experiment in mice. The human Tim-3-Ig fusion protein prepared by the present invention can be used for the treatment of immune disorders caused by the overexpression of Tim-3.
