The present invention relates to a method for culturing a mesenchymal stem cell, which comprises the steps of: a) coating a culturing container with gelatin; b) inserting a serum-free medium composition in the culturing container coated with gelatin; and c) inoculating and culturing the medium composition with a mesenchymal stem cell. In addition, the present invention relates to a manufacturing method of a medium for culturing a mesenchymal stem cell, which comprises the steps of: a) coating a culturing container with gelatin; and b) inserting a serum-free medium composition in the culturing container coated with gelatin. In the case of using a serum-independent medium disclosed in the present invention, a mesenchymal stem cell can be cultured in quantities without using animal serum such as fetal bovine serum or human serum.
