The present invention relates to a method for effectively analyzing an interaction between DNA and p53 protein by using a single plasmonic nanosensor. More particularly, the invention relates to a method for analyzing a difference of an interaction between a wild-type p53 protein and a mutant-type p53 protein both bonded to GADD45 promoter and a difference of a bonding force therebetween by using a sensor having a single gold nanoparticle fixed thereto. The analysis method by using a single plasmonic nanosensor of the present invention is capable of directly analyzing a difference of an interaction and a bonding force of the p53 protein bonded to a particular promoter without a separate labeling process, and capable of identifying importance of each part in accordance with amino acid mutation by comparing a difference of a bonding force between the wild-type p53 protein and the mutant-type p53 protein. Accordingly, the invention may be applied and used in the analysis field for diagnosing a patient with a high potential to develop a tumor at an early stage.
