The present invention relates to a method for preparing probiotics using vegetable mucin and water-soluble dietary fibers extracted and separated from Dioscorea opposite Thunb. When probiotics of edible microorganisms, such as lactic acid bacteria, are prepared, vegetable mucin separated from Dioscoreaceae crops and water-soluble dietary fibers widely commercialized as a food raw material as a freeze protectant and a micro-coating binding agent are added to a high-concentration of bacterial liquid collected after a microorganism culturing process or an existing probiotic powder obtained by freeze-drying the bacterial liquid, thereby increasing the production yield of probiotics and improving the acid resistance, bile resistance, and intestinal residence. Therefore, the present invention can maximize the physiological effect of probiotics when the probiotics are orally ingested, and can be useful in the process of preparing probiotics having enhanced viability during the distribution procedure.
