PROBLEM TO BE SOLVED: To provide an orthogonal translation system for producing a polypeptide incorporating unnatural amino acids in a methylotrophic yeast such as Pichia pastoris, as well as a method for producing polypeptides incorporating unnatural amino acids in a methylotrophic yeast such as Pichia pastoris.SOLUTION: Provided is a methylotrophic yeast cell comprising: a) an unnatural amino acid; b) an orthogonal aminoacyl synthetase (O-RS); and c) an orthogonal tRNA (O-tRNA) that recognizes the selector codon and is preferentially aminoacylated with the unnatural amino acid by the O-RS in a methylotrophic yeast cell.SELECTED DRAWING: Figure 6
