PROBLEM TO BE SOLVED: To provide a method for producing polymeric straight chain duplex DNA by promoting the binding between DNA molecules without circularizing a plurality of straight chain duplex DNAs within the same molecule.SOLUTION: The invention discloses a method for producing duplex DNA in which a plurality of straight chain duplex DNAs are linked in series by incubating the plurality of straight chain duplex DNAs with DNA ligase under the presence of RecA family recombinant enzyme or a protein with recombinant activity to inhibit the binding of both ends within the straight chain duplex DNA molecule, and to promote the binding between the ends of straight chain duplex DNA molecules.SELECTED DRAWING: None
