PROBLEM TO BE SOLVED: To provide heat-resistant glucose-6-phosphate dehydrogenase (G6 PDH) in which affinity to coenzyme NAD is improved.SOLUTION: A production method of the polypeptide comprises a polypeptide which has G6 PDH activity, and in which an amino acid residue at a specific position is substituted by other amino acid residue, and in which affinity to coenzyme NAD is improved as compared with the original G6 PDH; DNA encoding the polypeptide; a recombinant vector containing the DNA; a transformant containing the nine vectors; and steps of culturing transformant, in an amino acid sequence of heat-resistant G6 PDH derived from Geobacillus stearothermophilus.
