The invention relates to a polarization microscope using a polarization state of a spectrum-encoded cell. Polarization microscopes based on this spectral coding are fast enough for cell imaging and are compatible with optics with high extinction ratio characteristics that require imaging of molecular structures and molecular populations. The encoded polarization microscope allows for a null measurement of cell birefringence to be observed, while at the same time allowing the polarization state applied to the sample to sample individual discrete states throughout the Poincare sphere. Sampling the entire Poincare sphere makes it possible to determine the phase difference of the sample caused by linear and circular birefringence in the microscope. Polarization microscopes based on spectral coding can operate even in slightly non-null states, improving the signal-to-noise ratio. [Selection] Figure 1
