Generic method for determining the real-time PCR cycle threshold

in a standard sigmoidal amplification curves, and a single technique for determining the cycle threshold is provided in the PCR which may be used for amplification curves with problems such as parabolic curves. Ct values, the tangent to the curve at the maximum value of the second derivative can be measured as an intersection of the baseline of the amplification curve. Such Ct value may provide a sigmoidal curve and can be used for parabolic curve and linear correction for achieving accuracy in the measurement of the initial concentration of the sample.
