The present invention provides improved tissue culture Protocol for large scale production of Tectona grandis L., which is presently recognized as one of the valuable timber species in tropics. Said improved culture protocol saves time of development (at least 20% time) besides improved yield and quality of wood. . The process technique according to present invention also retains genetic purity of high standard. This protocol involves use of selected healthy mother plant nodal explants which are aseptically transferred on to an agarified SSWP initiation medium with 3% sucrose (HiMedia). Media was adjusted to pH 5.7 � 2 prior to autoclaving at 121 �C for 15 minutes. Subsequently-. the cultures were transferred to growth room with 7 days continuous dark its followed by 10 days 16/8 h light/dark cycle at about 27�2 �C temperatures. After 15-20 days of shoots sprouting, shoots are sub cultured on SSWP multiplication medium. The healthy multiplied shoots were transfer to SSWP rooting medium with about 0.5-1% activated charcoal. These cultures are kept under incubation for about 20-25 days under controlled conditions with 27�2 �C temperatures and with 16 hours light and 8.0 hours dark by providing and 3000 lux light intensity with the help of cool white florescent lights. .... Remove the plants having roots from bottle and dip the plants in bavistin (0.1%) + charcoal (0.5%) solution and kept for 10 min. than collect plants in Tupperware boxes. The rooted plants are transferred to portray, than next.the portrays were transfer to baby tunnel under controlled 33 �C temperature and 70% humidity for 20 to 25 days. Apply sufficient water with help of sprayer on 3rd, 6th and 9th day of incubation. Spray 5gm/101it N: P: K (12:61:00) on 10th day and 10 gm/lOlit N: P: K (19:19:19) on 14th day. Check the roots initiated on 18-23rd day and open baby tunnel.for 48hr. Apply the sufficient water to the primary hardened tray plants. Fill the pouches with soil, sand and organic manure with biofertilizers of 1:1:1 proportion. Transfer the plants in pouches by dipping entire plug in pouch and keeping two nodes above the soil level. Set the 33 �C temperature and 70% humidity. Put the pouches on beds and apply sufficient water to plants daily. On 10th day drench the plants with 50 ml NPK 19:19:19 (2 gm /lit cone.) and on 20th day apply 10 ppm micronutrient foliar spray. On 30th day apply 2 gm /lit 12:61:00 foliar spray and the pouched commercial plantlets are ready to dispatch for plantations, which not only save time of growth but also retains quality and quantity of wood.
