The present invention relates to use of a natural dye derivative for staining Nucleic acid derived from R. cordifolia dye (DSJP) - A safer, eco-friendly alternative to replace Ethidium bromide, Gel electrophoresis is a technique widely utilized to separate DNA fragments by their size or molecular weight. The agarose gel acts like a sieve, separating different sized fragments while the electric current provides the driving force. Later, these fragments or DNA bands are stained by some dye to visualize it. Ethidium bromide (3,8-diamino-5-ethyl-6-phenylphenanthridinium bromide) is the most commonly used nucleic acid (DNA, RNA) stain for gel electrophoresis because of low cost, ease of use, and the ability to detect DNA in submicrogram amounts. DSJP dye is prepared by derivatization of the product isolated from the natural dye extract of Rubia cordifolia. The natural dye extract was prepared from the methanolic extraction of stem and leaves of R. Cordifolia. Column chromatography of the product identified as Alizarin, was further derivatized by diallylation reaction to prepare DSJP. DSJP dye was found to be a safer alternative to replace Ethidium bromide, The IUPAC nomenclature and structure of the natural dye derivative is 2-[ 1 -(2-Methyl-allyloxy)-9,10-dioxo-9,10-dihydro-anthracen-2-yloxymethyl]-acrylic acid ethyl ester
