The present invention relates to an optimal diastatic enzyme mixture in accordance with a pretreatment method of lignocellulose, and to a manufacturing method thereof. The mixture and the method have effects in improving a saccharification rate by controlling a mixing rate and a composition suitable for an enzyme cocktail used for saccharification with respect to acid- or alkali-pretreated cellulose-based biomass. The composition for saccharifying lignocellulose comprises an enzyme cocktail mixed with endoglucanase derived from Saccharophagus degradans, cellobiohydrolase derived from Thermobifida fusca, and endoxylanase derived from Saccharophagus degradan, with respect to -glucosidase derived from Aspergillus niger.
