The present invention relates to a non-fluorescent detection method of a target biomolecule, and to a system for detecting a total internal reflection scattering signal for the same. Specifically, the present invention relates to: a detection method for detecting a scattering signal from a first metal-nanopattern and a second metal-containing nanoparticle, by using a nanobiochip including a first metal-nanopatterned substrate having scattering spectrums which do not overlap with each other, and by using a detection molecule tagged by the second metal-containing nanoparticle and specifically bonded to the target biomolecule; and a system for detecting a total internal reflection scattering signal for the same. The method is capable of effectively detecting a small amount of samples, by improving a signal-to-noise ratio (S/N) of the metal nanoparticle and lowering the detection limit.
